Psilocybin worked on anaesthetised mice

Only in the autism-model strain — and the "no trip needed" case still rests on rodents.

1. Can you get the effect without the trip?

A single psilocybin dose raised sociability in autism-model mice even when given under anaesthesia.

In Cntnap2-knockout mice the effect was durable; isogenic controls showed no change. Separately, a phase 1 study in 56 healthy adults mapped psilocybin from 0.5 to 4.0 mg, finding dose-proportional psilocin exposure, placebo-comparable adverse events and somnolence as the main signal — a defined subperceptual range.

2. Safety evidence came from surveys and case notes

This week's safety data are observational, and one case report documents psilocybin making things worse.

JAMA Network Open published a nationally representative US cross-sectional survey of health outcomes attributed to recent psilocybin, LSD, MDMA, ibogaine and 5-MeO-DMT use. A separate case report describes worsening depression and anxiety after psilocybin in cyclothymia, and an Oregon risk-reduction consult service (n=29) found most patients low-risk, with 17% reporting a prior challenging experience.

3. Anti-inflammatory in mice, equivocal in people

A meta-analysis of 61 studies finds the immune story holds preclinically and largely fails to replicate in humans.

Most in vitro (19/24) and non-human in vivo (25/30) studies reported anti-inflammatory effects, and pooled murine inflammatory models showed reduced proinflammatory cytokines. Of ten human trials, only three showed anti-inflammatory effects.

4. What is actually in the supply

One in six street samples in New South Wales carried an adulterant, and 3.9% contained a novel psychoactive substance.

Across 3,378 seized samples analysed 2021–2025, NPS detections included dipentylone, nitazenes, fentanyl analogues and novel benzodiazepines; median MDMA tablet dose was 104 mg and rising. Chinese forensic chemists separately characterised ZC-B, an azetidine analogue of 2C-B, in seized capsules with no library match.
